scenario: User1 is connected locally to the machine using his User1 account. User2 connects remotely with NoMachine to his User2 account.
Am I right to assume that the above is not possible with at least the free version?

Yes, your friend can connect to your computer whilst you are sitting physically in front of it. Please check this article here: https://www.nomachine.com/AR10K00716. This is considered one remote connection.
What is not possible is if another person connects to your desktop remotely whilst you are also connect remotely. This would be considered two connections.

You, user1, are sitting in front of the computer logged in to “Britgirl” account. I, user2, connect to the computer remotely to “Loke” account.
I tried it and it just connected me to the already running user1 account. Would this be possible via the virtual desktops and what not?
If you want to have another user logged in to your Linux desktop whilst you are working on it locally (you are sat in front of it), you can install the free version of NoMachine and your friend can connect from remote.
If you want to do this with virtual desktops, you need the Workstation product: https://www.nomachine.com/product&p=NoMachine%20Workstation. The virtual desktop functionality is a feature of the Terminal Server family.

https://www.nomachine.com/AR10K00702Wow well that’s way outside my budget range. I wouldn’t mind paying a small price since I use NoMachine a lot but for personal use (using it to connect to my university pc) that is way too much.

The Workstation (as with all the Terminal Server products) work as a terminal server, by creating sessions running in the background. These are terminal servers, intended for the enterprise market. There are single licenses available for Workstation (maybe you saw the price of the pack of 10) and there are often ongoing promotions, especially for universities (why not contact the sales team to ask for more info?). What you should do is try the evaluation software which is freely available and see if it fits your requirements.
